چکیده

Background Current treatments of chemotherapy-induced cardiomyopathy (CCM) are limited efficacy. We assessed whether repeated intravenous injections human extracellular vesicles from cardiac progenitor cells (EV-CPC) could represent a new therapeutic option and EV manufacturing according to Good Manufacturing Practices (GMP)-compatible process did not impair their bioactivity. Methods Immuno-competent mice received intra-peritoneal (IP) doxorubicin (DOX) (4 mg/kg each; cumulative dose: 12 mg/kg) were then intravenously (IV) injected three times with EV-CPC (total 30 billion). Cardiac function was 9–11 weeks later by magnetic resonance imaging (CMR) using strain as the primary end point. Then, immuno-competent rats 5 IP DOX (3 dose 15 followed 3 equal IV GMP-EV 100 two dimensional-echocardiography. Results In chronic mouse model CCM, + placebo-injected hearts incurred significant decline in basal (global, epi- endocardial) circumferential compared sham DOX-untreated ( p = 0.043, 0.042, 0.048 respectively) while preserved these indices. Global longitudinal similar pattern. rat model, also left ventricular end-systolic end-diastolic volumes untreated controls. Conclusions Intravenously-injected derived CPC have cardio-protective effects which may make them an attractive user-friendly for treatment CCM.

Therapeutic Potential of Extracellular Vesicles

Extracellular vesicles (EV) have emerged as important mediators of intercellular communication. By their origin, we can find vesicles derived from plasma membrane such as microvesicles, ectosomes, and membrane particles or exosomes, which originate in the endosomal membrane compartment. They contain numerous proteins, lipids, and even nucleic acids like mRNA and miRNA that can affect the cells ...

Focus on Extracellular Vesicles: Therapeutic Potential of Stem Cell-Derived Extracellular Vesicles

The intense research focus on stem and progenitor cells could be attributed to their differentiation potential to generate new cells to replace diseased or lost cells in many highly intractable degenerative diseases, such as Alzheimer disease, multiple sclerosis, and heart diseases. However, experimental and clinical studies have increasingly attributed the therapeutic efficacy of these cells t...

Characterization of extracellular vesicles derived from cardiac cells in an in vitro model of preconditioning

Preconditioning is a promising technique to protect the heart from ischaemia-reperfusion injury. In this context, the crosstalk between different cardiac cell types and especially the exchange of cardioprotective mediators has come into the focus of current research. Recently, extracellular vesicles (EVs), nano-sized structures, emerged as possible communication mediators. They are taken up by ...

Mesenchymal Stem Cell–Derived Small Extracellular Vesicles Promote Neuroprotection in Rodent Models of Glaucoma

Purpose To investigate the benefit of bone marrow mesenchymal stem cell (BMSC)-derived small extracellular vesicles (sEV) as an intravitreal (ivit) therapy in two rat models of glaucoma and to determine and identify candidate miRNA involved in the mechanism. Methods sEV were isolated from human BMSC and fibroblasts and ivit injected into adult rats after induction of elevated IOP. IOP was ele...
